The conserved Mg2+-dependent phosphatidic acid phosphatase, as depicted by the predicted structure of its catalytic core (N-LIP domain, bronze; HAD-like domain, cyan, and amphipathic helix, magenta), catalyzes the dephosphorylation of phosphatidic acid to produce diacylglycerol, a key step in lipid metabolism. Yeast Pah1 and human lipin 1 forms of the enzyme are targeted for inhibition at the catalytic core by the antidepressant drug sertraline.
