Why do studies claiming #Treg induction by novel designer drugs test them only when given before or with antigen, rather than in established disease that more closely reflects human cases?

The identification of regulatory T cells and #Foxp3 has shaped modern #immunology and contributes to antibody-research approaches worldwide! Along with spurring the advancement in treatments for #cancer and #AutoimmuneDiseases.
Congrats to Drs. Mary Brunkow and Fred Ramsdell on receiving the #NobelPrize in Physiology or Medicine! 🏆
We discuss their early work identifying #FOXP3 as a master regulator of regulatory T cells.

Mary Brunkow’s #FOXP3 discovery earned her a Nobel, though she left the field soon afterward. Douglas Prasher cloned #GFP in 1992, left the field too, and was notably left out of the #Nobel for its discovery.

The discovery of #FOXP3 was the key that unlocked the puzzle.
They found that without the FOXP3 gene, regulatory T cells cannot form. This allows the #ImmuneSystem to go wild, attacking the body's own organs, a condition mirrored in a rare human disease called #IPEX.

But what controlled these crucial cells? That answer came from Drs. Brunkow and Ramsdell.
After years of arduous work at a biotech company, they studied mice with a severe #autoimmune disease and found the single missing gene responsible. They named it #FOXP3.

Sakaguchi entdeckte die regulatorischen T-Zellen #Treg, Brunkow und Ramsdell den dazugehörigen Transkriptionsfaktor #FoxP3.
Große Freude, weil jahrelang mein täglich Brot.

Great recognition to a fundamental discovery that reshaped modern #immunology. Congrats to the laureates!
However, imho the Nobel committee missed the chance to acknowledge an additional key contributor to the understanding of #Foxp3 and #Treg functionality

Not just ‘memory T cells act as microbial surrogates to drive innate immune activation and inflammation’, activated CD4 T cells can as well. This is precisely why only antigen-specific Tregs can keep anti-self T cell responses from spiraling out.

ONLINE COVERLNP mRNA Vaccines Get a Boost. Lipid nanoparticle (LNP) mRNA vaccines induce robust immune responses and provide protection against infectious diseases, but this immunity can be short-lived, necessitating frequent booster vaccinations. Aunins et al. investigated how adjuvanting LNP-mRNA vaccines with mRNA encoding the cytokine interleukin-12 (IL-12) influences CD8 T cell responses in mice. Il12 mRNA-LNPs (LNP–IL-12) enhanced antigen-specific CD8 T cell responses and CD8 T cell memory, providing greater protection against bacterial infection and in a mouse model of melanoma. Cytokine-adjuvanted mRNA-LNPs may help future vaccine platforms better tailor the magnitude and nature of the immune response. This month’s cover image features a stylized mRNA-LNP with various lipid (red, coral, and pink) and cholesterol molecules (maroon) encapsulating Il12 mRNA (yellow). Credit: A.
