Researchers Identify Protein Interaction Driving Alzheimer’s and Develop Compound to Halt It in Mice A research team led by Professor Hilmar Bading at Heidelberg University has uncovered a key biological mechanism behind Alzheimer's disease progression. The scientists identified a harmful interaction between two proteins—the NMDA receptor and the TRPM4 ion channel—that, when bound together, form a destructive protein complex. This complex triggers neuronal death and accelerates memory loss. Using a specially developed compound called FP802, the researchers successfully disrupted this toxic interaction in a mouse model of Alzheimer’s. The drug, described as a TwinF Interface Inhibitor, binds to the exact site where the two proteins connect, preventing the formation of the damaging complex. Mice treated with FP802 showed slower disease progression, preserved memory and learning abilities, and reduced brain damage. The treatment also lowered the accumulation of beta-amyloid plaques, a hallmark of Alzheimer’s pathology. Unlike traditional therapies focused solely on amyloid removal, this approach targets the downstream molecular mechanism responsible for neuronal death. The research suggests that FP802 may also have potential in treating other neurodegenerative diseases, such as ALS, that involve the same protein interaction. Although still in early preclinical stages, the discovery opens a promising path toward developing new therapies that protect brain cells and halt disease progression at its root. The team, collaborating with FundaMental Pharma, plans to refine FP802 and conduct safety and pharmacological studies before human trials.

Researchers Develop Light-Controlled Protein Evolution for Dynamic Cellular Functions Scientists at the Ecole Polytechnique Fédérale de Lausanne have developed a novel method called 'optovolution' that uses light to guide the evolution of proteins with dynamic behaviors.
