Enrichment of evolutionary young fixed TE copies near multi-ethnic TE-GWAS hits

Juan next hypothesized that SNVs 🧬 may reside in #TE copies that could provide substrates for de novo #transposition or #homology-dependent structural remodeling (think #NAHR). Consistently, he observed an enrichment of #evolutionarily young #LINE1 copies and intermediate-aged #Alu copies. 6/8

Significant enrichment for known TE regulators

When analyzing genes in LD with SNVs 🧬 🖥️ , we observed an enrichment of genes regulating #LINE1 expression, suggesting that our approach could capture bona fide #TE regulators, and that these regulators may be at play across #diverse human 👫 populations. 5/8

Manhattan plot shows significant genome-wide associations of genetic variants with the likelihood to carry unique LINE1/Alu insertions.

To our surprise, Juan identified many loci with #genome-wide significance, suggesting these polymorphic SNVs/SVs may influence #LINE1 & #Alu transposition! Interestingly, one SV was an inversion within the APP gene, a marker for #Alzheimers 🧠, reinforcing the links between #TEs and disease. 4/8
