Day 7: #glucagon has been used for beta blocker toxicity (unlicensed). Mechanism is increased cAMP in myocardium by-passing the beta-adrenergic messaging system; this exerts inotropic & chronotropic action. Also has been used in CCB overdose with similar effects. Evidence base unclear

Day 6: Drug-drug interactions. #glucagon can increase the anti-coagulant effect of warfarin & this is classified as ‘severe’ in BNF. Antagonised by insulin & it antagonises beta blockers > tachycardia & increased BP (short effect reflecting short t½). Action is compromised by indomethacin

Day 4: #glucagon levels rise when glucose levels drop too low. It binds glucagon receptors (mainly liver) & via secondary signals activates protein kinase A (PKA). This signals the liver to release stored glucose & to up-regulate new glucose production thus returning blood sugar to normal range

Day 3: #glucagon is relatively large & mainly stays in plasma/ECF so a low Vd. Minimal bioavailability differences between injection routes. Metabolised by protein degradation in plasma, liver, kidney & eliminated by latter two. Short half-life ~ 25-35 mins

Day 2 (cont). #glucagon is also used to inhibit gut motility as a diagnostic aid in radiological procedures

Day 2: #glucagon is licensed for diabetic hypoglycaemia for those over 4 weeks old by s.c, i.m, i.v routes. Dose 1mg for > 9 yrs. 500micrograms 1 month-8yrs. Used as unlicensed for cardio effects from acute overdose of beta blockers

Day 1: Identified from animal pancreatic extracts in 1920s (post insulin discovery), #glucagon is a peptide hormone named for ‘gluc’ose raising & ‘agon’ for agonist action > glucose release. Available as injection since 1960s & as synthetic version in pre-filled pen for use outside clinical setting
