Formation and dynamics of the plasma membrane reservoir (PMR). The PMR forms prior to the development of the daughter cells’ IMC. Its position and size vary during replication, and it is reabsorbed at the conclusion of daughter cell budding. We propose that the PMR facilitates the expansion of the PM surface during replication. All scale bars represent 1 µm. While the PMR is not always visible due to its transient nature, virtually all the parasites undergoing replication are forming a PMR.

We have a #SuperResolution #SMLM image of #mitochondria (magenta) and #lysosomes (yellow), and a diffraction-limited image of the #nucleus (blue). Next, we have the #cytoskeleton in green, alone or with the #PlasmaMembrane in gray. All layers can be generated with our user-friendly software, CODI.
Fixed Cos7 cells were fixed and stained in 5 colors:
🔬 #SMLM – The mitochondrial protein #Tomm20 (yellow) and the lysosomal marker #LAMP11 (red).
🔬 In diffraction-limited imaging – #Tubulin (cyan), the nuclear marker Hoechst (#DNA, magenta), and the #PlasmaMembrane dye CellMask™ Green (gray).
In addition, we have 3 diffraction-limited channels that provide a wider cellular context, namely, the #PlasmaMembrane, #Nucleus and the #microtubule #cytoskeleton, all used to facilitate accurate cellular orientation.
